    Memory and cognition in schizophrenia.

    Episodic memory deficits are consistently documented as a core aspect of cognitive dysfunction in schizophrenia patients, present from the onset of the illness and strongly associated with functional disability. Over the past decade, research using approaches from experimental cognitive neuroscience revealed disproportionate episodic memory impairments in schizophrenia (Sz) under high cognitive demand relational encoding conditions and relatively unimpaired performance under item-specific encoding conditions. These specific deficits in component processes of episodic memory reflect impaired activation and connectivity within specific elements of frontal-medial temporal lobe circuits, with a central role for the dorsolateral prefrontal cortex (DLPFC), relatively intact function of ventrolateral prefrontal cortex and variable results in the hippocampus. We propose that memory deficits can be understood within the broader context of cognitive deficits in Sz, where impaired DLPFC-related cognitive control has a broad impact across multiple cognitive domains. The therapeutic implications of these findings are discussed

    Triple-Mode Cavity Bandpass Filter on Doublet with Controllable Transmission Zeros

On the basis of doublet and its properties, a class of multiple-mode narrow band bandpasss filter is designed and fabricated by simultaneously exploiting the three resonant modes in a single rectangular cavity: TE101, TE011, and TM110 modes. The input/output ports of the proposed filter are fed by coupling a microstrip line to a slot on the side wall of a rectangular cavity. Different modes are excited by changing the position and shape of the two slots at input and output of the rectangular cavity without any intra-cavity coupling. Besides three poles within the passband, a pair of transmission zeros (TZs) is achieved, which can be controlled independently by setting the positions of the two TZs at the lower and/or upper stopband. High stopband attenuation and high filtering selectivity are achieved by considerably allocating three transmission poles and two zeros.     Wideband Filtering Out-of-Phase Power Dividers Using Slotline Resonators and Microstrip-to-Slotline Transitions

    © 2019 IEEE. Two wideband filtering out-of-phase power dividers are designed and analyzed. The initial design is composed of a quarter-wavelength slotline resonator and two microstrip-to-slotline transitions. Three transmission poles are produced to form the passband. Excellent matching and isolation are achieved using extra matching network. Based on the initial design, a pair of stepped-impedance open stubs are shunted at the output ports to improve the passband selectivity. The designs have been verified through experiment. The tested results show that the proposed devices have achieved 60% operating bandwidth, 1.8 dB insertion loss, 1.2° phase error and excellent in-band matching and isolation

    Targeting quiescent leukemic stem cells using second generation autophagy inhibitors

    In chronic myeloid leukemia (CML), tyrosine kinase inhibitor (TKI) treatment induces autophagy that promotes survival and TKI-resistance in leukemic stem cells (LSCs). In clinical studies hydroxychloroquine (HCQ), the only clinically approved autophagy inhibitor, does not consistently inhibit autophagy in cancer patients, so more potent autophagy inhibitors are needed. We generated a murine model of CML in which autophagic flux can be measured in bone marrow-located LSCs. In parallel, we use cell division tracing, phenotyping of primary CML cells, and a robust xenotransplantation model of human CML, to investigate the effect of Lys05, a highly potent lysosomotropic agent, and PIK-III, a selective inhibitor of VPS34, on the survival and function of LSCs. We demonstrate that long-term haematopoietic stem cells (LT-HSCs: Lin−Sca-1+c-kit+CD48−CD150+) isolated from leukemic mice have higher basal autophagy levels compared with non-leukemic LT-HSCs and more mature leukemic cells. Additionally, we present that while HCQ is ineffective, Lys05-mediated autophagy inhibition reduces LSCs quiescence and drives myeloid cell expansion. Furthermore, Lys05 and PIK-III reduced the number of primary CML LSCs and target xenografted LSCs when used in combination with TKI treatment, providing a strong rationale for clinical use of second generation autophagy inhibitors as a novel treatment for CML patients with LSC persistence
